El peróxido de hidrógeno reacciona con el agente cromógeno para producir una sustancia roja, que presenta un fuerte pico de absorción a 505 nm. En un rango de concentración determinado, su absorbancia es proporcional a la concentración de glucosa. Por lo tanto, la actividad de la sacarasa puede calcularse midiendo el valor de DO a 505 nm.
