COL4A3 es un componente estructural importante de las membranas basales. Está compuesto por tres subunidades alfa, codificadas por seis genes diferentes, del alfa 1 al alfa 6. Cada una de estas subunidades alfa puede formar una estructura de triple hélice con otras dos subunidades para formar COL4A3. La unión de autoanticuerpos a las moléculas de colágeno en las membranas basales de los alvéolos y glomérulos puede causar el síndrome de Goodpasture. COL4A3 también está vinculado a una forma autosómica recesiva del síndrome de Alport. COL4A3 presenta una conformación cabeza a cabeza y cada par de genes comparte un promotor común.
