El HGF es una proteína pleiotrópica de la subfamilia del plasminógeno de las peptidasas S1. El HGF murino se secreta como un propéptido monocatenario inactivo de 728 aminoácidos (aa). Una serina proteasa lo escinde después del cuarto dominio Kringle para formar un HGF bioactivo unido por enlaces disulfuro con una cadena alfa de 60 kDa y una cadena beta de 30 kDa. El HGF se une a los proteoglicanos de heparán-sulfato y al receptor de tirosina quinasa, ampliamente expresado, HGF R/c-MET. El HGF regula la morfogénesis epitelial induciendo la dispersión celular y la tubulogénesis ramificada. También puede alterar la morfología del epitelio induciendo la liberación del ectodominio alfa de la nectina-1, un componente de la proteína de adhesión de las uniones adherentes.
