PPP1R14A se une directamente a la proteína quinasa C y a la caseína quinasa I. PPP1R14A es un inhibidor dependiente de la fosforilación de la miosina fosfatasa del músculo liso. PPP1R14A es el inhibidor de PPP1CA. Cuando se fosforila, PPP1R14A presenta una actividad inhibitoria más de 1000 veces mayor, creando un interruptor molecular para regular el estado de fosforilación de los sustratos de PPP1CA y la contracción del músculo liso. Además, la inhibición de PPP1R14A también mejora la contracción del músculo liso en ausencia de un aumento de la concentración intracelular de Ca₂.
