FAS es un receptor que contiene tres repeticiones TNFR-Cys y un dominio de muerte. Se ha demostrado que FAS participa en la regulación fisiológica de la muerte celular programada y se ha relacionado con la patogénesis de diversas neoplasias malignas y enfermedades del sistema inmunitario. FADD (molécula adaptadora) recluta la caspasa-8 hacia el receptor activado; el complejo de señalización inductor de muerte (DISC) resultante realiza la activación proteolítica de la caspasa-8, lo que inicia la cascada subsiguiente de caspasas que median la apoptosis. La apoptosis mediada por FAS puede desempeñar un papel en la inducción de tolerancia periférica, en el suicidio de células T maduras estimulado por antígenos, o en ambos.
