En presencia de la peroxidasa, el peróxido de hidrógeno se oxida para producir el sustrato de fluorescencia. La intensidad de la fluorescencia a una longitud de onda de excitación de 535 nm y una longitud de onda de emisión de 587 nm es proporcional al contenido de glucógeno.
