La escisión de la α-secretasa impide la formación de Aβ, mientras que las escisiones β- y γ-secretasa generan componentes de la APP con características amiloidogénicas. La proteína precursora de beta amiloide A4 (APP), codificada por el gen APP, localizado en el cromosoma humano 21q, es un receptor de superficie celular y desempeña funciones fisiológicas en la superficie de las neuronas, relevantes para el crecimiento de neuritas, la adhesión neuronal y la axonogénesis. La APP se expresa en todos los tejidos fetales y es pronunciada en el cerebro, riñón, corazón y bazo, pero débil en el hígado. Los defectos en la APP son la causa de la enfermedad de Alzheimer tipo 1 (EA1).
